No, it will have to be NAT port you have been assigned.
You'd have to connect via 1 of your 20 assigned ports. For example one of my boxes has 17901-17920.

Even the assigned ports might not work over IPv4, because a TCP header is required for the NAT to work.

They do work. Anyway, OpenVPN can be used over TCP too.
As already mentioned: you can use port 443 if you can connect over ipv6 not over ipv4.
If you need to connect using ipv4 you need to configure openvpn to use one of the assigned 20 ports.
That port will also be available uskng ipv6.
